ÿþ<!DOCTYPE html PUBLIC "-//W3C//DTD XHTML 1.0 Transitional//EN" "http://www.w3.org/TR/xhtml1/DTD/xhtml1-transitional.dtd"> <html lang="fr"> <head> <meta http-equiv="Content-Type" content="text/html; charset=utf-8" /> <title>Homepage of Alain Chaumont (Contact)</title> <link rel="stylesheet" type="text/css" href="stylesheet.css"> </head> <body> <div id="global"> <div id="header"> </div> <div id="center"> <div id="content"> <br> <br> <h2> hGBP1 </h2> <img src="pics/hgbp1.jpeg" width="263" height="350" border="0" vspace="3" hspace="15" align="left"> <p> The human guanylate binding protein 1 (hGBP1), who belongs to the family of dynamin-related large GTP-binding proteins, has been shown to provide resistance against the proliferation of the vesicular stomatitis and encephalomyocarditis viruses. </p> <p>This family of dynamin-related large GTP-binding proteins share common biochemical properties which are not found in other families of GTP-binding proteins, such as nucleotide-dependent oligomerization and fast cooperative GTPase activity. However one special feature of hGBP1 is that it hydrolysis GTP to GMP in two consecutive cleavage reactions.</p> <p> The catalytic machinery as well as the contribution of the different domains to the GTPase reaction remain elusive, and conflicting models have been presented for the role of the C-terminal GTPase effector domain of dynamin, which retains the main biochemical properties of hGBP1 such as nucleotide affinity, GTP-induced homodimerization, cooperative GTPase activity. </p> <p> Goal of this project is to study the different mechanism of the GTP to GDP and GDP to GMP cleavage reactions using computational chemistry tools.
